Structural insight into African horsesickness virus infection
African horsesickness (AHS) is a devastating disease of horses. The disease is caused by the double-stranded RNA-containing African horsesickness virus (AHSV). Using electron cryomicroscopy and three-dimensional image reconstruction, we determined the architecture of an AHSV serotype 4 (AHSV-4) reference strain. The structure revealed triple-layered AHS virions enclosing the segmented genome and transcriptase complex. The innermost protein layer contains 120 copies of VP3, with the viral polymerase, capping enzyme, and helicase attached to the inner surface of the VP3 layer on the 5-fold axis, surrounded by double-stranded RNA. VP7 trimers form a second, T 13 layer on top of VP3. Comparative analyses of the structures of bluetongue virus and AHSV-4 confirmed that VP5 trimers form globular domains and VP2 trimers form triskelions, on the virion surface. We also identified an AHSV-7 strain with a truncated VP2 protein (AHSV-7 tVP2) which outgrows AHSV-4 in culture. Comparison of AHSV-7 tVP2 to bluetongue virus and AHSV-4 allowed mapping of two domains in AHSV-4 VP2, and one in bluetongue virus VP2, that are important in infection. We also revealed a protein plugging the 5-fold vertices in AHSV-4. These results shed light on virus-host interactions in an economically important orbivirus to help the informed design of new vaccines
How Supervisors Influence Performance: A Multilevel Study of Coaching and Group Management in Technology-Mediated Services
This multilevel study examines the role of supervisors in improving employee performance through the use of coaching and group management practices. It examines the individual and synergistic effects of these management practices. The research subjects are call center agents in highly standardized jobs, and the organizational context is one in which calls, or task assignments, are randomly distributed via automated technology, providing a quasi-experimental approach in a real-world context. Results show that the amount of coaching that an employee received each month predicted objective performance improvements over time. Moreover, workers exhibited higher performance where their supervisor emphasized group assignments and group incentives and where technology was more automated. Finally, the positive relationship between coaching and performance was stronger where supervisors made greater use of group incentives, where technology was less automated, and where technological changes were less frequent. Implications and potential limitations of the present study are discussed

Dynamic Adaptation of Software-defined Networks for IoT Systems: A Search-based Approach
The concept of Internet of Things (IoT) has led to the development of many complex and critical systems such as smart emergency management systems. IoT-enabled applications typically depend on a communication network for transmitting large volumes of data in unpredictable and changing environments. These networks are prone to congestion when there is a burst in demand, e.g., as an emergency situation is unfolding, and therefore rely on configurable software-defined networks (SDN). In this paper, we propose a dynamic adaptive SDN configuration approach for IoT systems. The approach enables resolving congestion in real time while minimizing network utilization, data transmission delays and adaptation costs. Our approach builds on existing work in dynamic adaptive search-based software engineering (SBSE) to reconfigure an SDN while simultaneously ensuring multiple quality of service criteria. We evaluate our approach on an industrial national emergency management system, which is aimed at detecting disasters and emergencies, and facilitating recovery and rescue operations by providing first responders with a reliable communication infrastructure. Our results indicate that (1) our approach is able to efficiently and effectively adapt an SDN to dynamically resolve congestion, and (2) compared to two baseline data forwarding algorithms that are static and non-adaptive, our approach increases data transmission rate by a factor of at least 3 and decreases data loss by at least 70%
Religion and religious education : comparing and contrasting pupilsâ and teachersâ views in an English school
This publication builds on and develops the English findings of the qualitative study of European teenagersâ perspectives on religion and religious education (Knauth et al. 2008), part of âReligion in Education: A contribution to dialogue or a factor of conflict in transforming societies of European countries?â (REDCo) project. It uses data gathered from 27 pupils, aged 15-16, from a school in a multicultural Northern town in England and compares those findings with data gathered from ten teachers in the humanities faculty of the same school, collected during research for the Warwick REDCo Community of Practice. Comparisons are drawn between the teachersâ and their pupilsâ attitudes and values using the same structure as the European study: personal views and experiences of religion, the social dimension of religion, and religious education in school. The discussion offers an analysis of the similarities and differences in worldviews and beliefs which emerged. These include religious commitment/observance differences between the mainly Muslim-heritage pupils and their mainly non-practising Christian-heritage teachers. The research should inform the ways in which the statutory duties to promote community cohesion and equalities can be implemented in schools. It should also facilitate intercultural and interreligious understanding between teachers and the pupils from different ethnic and religious backgrounds
Education policy as an act of white supremacy: whiteness, critical race theory and education reform
The paper presents an empirical analysis of education policy in England that is informed by recent developments in US critical theory. In particular, I draw on âwhiteness studiesâ and the application of Critical Race Theory (CRT). These perspectives offer a new and radical way of conceptualising the role of racism in education. Although the US literature has paid little or no regard to issues outside North America, I argue that a similar understanding of racism (as a multifaceted, deeply embedded, often taken-for-granted aspect of power relations) lies at the heart of recent attempts to understand institutional racism in the UK. Having set out the conceptual terrain in the first half of the paper, I then apply this approach to recent changes in the English education system to reveal the central role accorded the defence (and extension) of race inequity. Finally, the paper touches on the question of racism and intentionality: although race inequity may not be a planned and deliberate goal of education policy neither is it accidental. The patterning of racial advantage and inequity is structured in domination and its continuation represents a form of tacit intentionality on the part of white powerholders and policy makers. It is in this sense that education policy is an act of white supremacy. Following others in the CRT tradition, therefore, the paperâs analysis concludes that the most dangerous form of âwhite supremacyâ is not the obvious and extreme fascistic posturing of small neonazi groups, but rather the taken-for-granted routine privileging of white interests that goes unremarked in the political mainstream

Needle & knot : binder boilerplate tied up
To lighten the burden of programming language mechanization, many approaches have been developed that tackle the substantial boilerplate which arises from variable binders. Unfortunately, the existing approaches are limited in scope. They typically do not support complex binding forms (such as multi-binders) that arise in more advanced languages, or they do not tackle the boilerplate due to mentioning variables and binders in relations. As a consequence, the human mechanizer is still unnecessarily burdened with binder boilerplate and discouraged from taking on richer languages.
This paper presents Knot, a new approach that substantially extends the support for binder boilerplate. Knot is a highly expressive language for natural and concise specification of syntax with binders. Its meta-theory constructively guarantees the coverage of a considerable amount of binder boilerplate for well-formed specifications, including that for well-scoping of terms and context lookups. Knot also comes with a code generator, Needle, that specializes the generic boilerplate for convenient embedding in COQ and provides a tactic library for automatically discharging proof obligations that frequently come up in proofs of weakening and substitution lemmas of type-systems.
Our evaluation shows, that Needle & Knot significantly reduce the size of language mechanizations (by 40% in our case study). Moreover, as far as we know, Knot enables the most concise mechanization of the POPLmark Challenge (1a + 2a) and is two-thirds the size of the next smallest. Finally, Knot allows us to mechanize for instance dependentlytyped languages, which is notoriously challenging because of dependent contexts and mutually-recursive sorts with variables
Paediatric non-progression following grandmother-to-child HIV transmission
Background
In contrast to adult HIV infection, where slow disease progression is strongly linked to immune control of HIV mediated by protective HLA class I molecules such as HLA-B*81:01, the mechanisms by which a minority of HIV-infected children maintain normal-for-age CD4 counts and remain clinically healthy appear to be HLA class I-independent and are largely unknown. To better understand these mechanisms, we here studied a HIV-infected South African female, who remained a non-progressor throughout childhood.
Results
Phylogenetic analysis of viral sequences in the HIV-infected family members, together with the history of grand-maternal breast-feeding, indicated that, unusually, the non-progressor child had been infected via grandmother-to-child transmission. Although HLA-B*81:01 was expressed by both grandmother and grand-daughter, autologous virus in each subject encoded an escape mutation L188F within the immunodominant HLA-B*81:01-restricted Gag-specific epitope TL9 (TPQDLNTML, Gag 180â188). Since the transmitted virus can influence paediatric and adult HIV disease progression, we investigated the impact of the L188F mutant on replicative capacity. When this variant was introduced into three distinct HIV clones in vitro, viral replicative capacity was abrogated altogether. However, a virus constructed using the gag sequence of the non-progressor child replicated as efficiently as wildtype virus.
Conclusion
These findings suggest alternative sequences of events: the transmission of the uncompensated low fitness L188F to both children, potentially contributing to slow progression in both, consistent with previous studies indicating that disease progression in children can be influenced by the replicative capacity of the transmitted virus; or the transmission of fully compensated virus, and slow progression here principally the result of HLA-independent host-specific factors, yet to be defined
Class in contemporary Britain: comparing the Cultural Capital and Social Exclusion (CCSE) project and the Great British Class Survey (GBCS)
The paper discusses the salience of class in Britain in relation to the experiment of the BBCâacademic partnership of the Great British Class Survey (GBCS). It addresses the claimed inauguration of a third phase in class analysis in the UK sparked by the experiment. This is done by considering three main issues. First, the GBCS experiment is situated in the context of various explorations of cultural class analyses, and chiefly in relation to the Cultural Capital and Social Exclusion (CCSE) project (ESRC funded 2003â6). Secondly, the focus is on the influence of the academic turn to big data for the procedures and claims of the project, and some implications of the methodological choices. Thirdly, attention is turned to the deleterious effects of commercial and institutional pressures on the current research culture in which the experiment exists
